I finally got my butt in gear...

I cleaned up my office and put a lot of stuff away so that it finally looks like a livable room. I still have a lot of things to do, like put away the nick nacks and hang pictures and curtains, but everyday it looks more like an actual apartment that has someone living in it.

I had the day off today because my reagents hadn't come yet but they should be there tomorrow and I can get my experiment going.

I realized that I hadn't posted about my dinner with my fellow graduate students. We had a bbq at Amanda and Erin's apartment. Amanda, Christina, and Kevin are in my program and Erin is in the Flex program, which means that she hasn't decided exactly which program she wants to be in right now. They are all very nice. We have a lot in common (you know the whole being science geeks) and spent the night watching CSI and baseball. I like baseball but I don't understand it as much as I do football and basketball. I think that now that I live in a town with a major baseball team I need to learn a little bit more. At least enough that I don't make myself look like an idiot. I do wish that they liked philosophy a little bit more and I have yet to broach the subjects of politics and religion. I think I will get to know them a little bit more before I see what they believe.

Kathy thinks I should ask questions on my blog to see people's responses. I agree, it can be a little game to spice things up a little bit. The problem is coming up with questions to ask. I could ask a chemistry question or history question. Or I could ask personal questions. I already know what Kathy would prefer.

My first question is: Who was the last person you couldn't take your eyes off of?
I will give my answer after I get at least one answer. It is a give and receive type of relationship.

I finally read it....

I have finished Harry Potter (thanks to Dave for letting me borrow his copy). I think that I might actually get my own copy from Amazon soon, but I am not holding my breath. It is no longer important.

I feel myself getting on the verge of being sappy or melodramatic so I am going to bed, but I promise to do an actual update soon.

I want to give a shout out to the RHI Sports Program and the wonderful job they did over the weekend to raise money for the program. The last count for the earnings of Wheels in Motion was $13,000! It was a lot of fun too. It kinda makes me want a motorcycle but I am willing to wait. I want my advanced scuba diving license first.

Back and Forth Back and Forth...

I am going back to Indy again this weekend. I am going for the express reason of helping Janeen with a RHI Sports Program volunteering oppurtunity. If anyone is interested, it is tomorrow from basically 2-8 pm in Fishers. You would have to call me or Janeen for details because right now I don't know them. I am about to head out so I will be free to talk for 2 hours while I drive.

I am very frustrated with work because nothing is working out the way it is suppose to but hopefully since I ordered four new reagents today it will work next week. Cross my fingers.

Willy Wonka..Willy Wonka..Willy Wonka...

I loved the movie. For those who saw the movie-the title of the blog should be put to the music from the puppets (poor puppets). I love the fact that I have an IMAX theater ten minuets from my house. It was so wonderful to see that movie really big. I thought it was a great movie and just goes to show what a wonderful actor Johnny Depp is. The original will always hold a place in my heart, but this version made me laugh so hard. The Oompa Loompas were great-I loved their songs. I really liked the glass elevator, I wish I had one. It would make the trip to Indy a lot faster.

I have to go because I am at work but I just thought that I would mention before I go that Harry Potter and the Goblet of Fire will be release on IMAX at my theater. I will let people know when, time, and how much when I find out. I am really hoping that they will do a midnight showing on the IMAX, but I don't know if they will.

My plan back fired...

So I went back to Indy to pick up my Harry Potter book and guess what-it never came. Somewhere between Amazon and UPS it never got delivered. I am going to call UPS today to try and find out what happened. The plus side is that since I did not get the book on July 16, Amazon refunded my money. So now all I need is the actual book.

So, the short of it all is no I haven't read the book, and yes if anyone tells me what happens, I will be very angry. Physical harm angry.

The plus side to everything is that I am seeing Charlie and the Chocolate Factory today on IMAX. Oompa Loompas never looked so tall. That is of course if I can ever leave work. I am waiting for my PI to look over some slides so that I can separate some mice and go home. My head feels like someone stuffed cotton inside it and then took a baseball bat to it for a couple of hours. Don't you just love summer allergies?

Ok, enough complaining. I really am having fun here in Cinci and I will be back into Indy next weekend to help Janeen out with some RHI sports program volunteering (which if anyone is interested in helping, I am sure Janeen would love more help).

I just can't wait...

The new Harry Potter comes out on Saturday!!!! I am having it sent from Amazon since I know that way I will definitley get it on Saturday and won't have to deal with going to the stores.
If you haven't read the series yet, you are missing out. The movies are really good but the books are excellent. You get so much more about the characters from the books. It is almost like the movies are trailers for the books.

Not much happening. I hope something does happen soon or my blog is going to be permantely boring and no one will read it anymore. I did get car, renters, and identity theft insurance today. Ha, now no one can still my identity. Though I don't know why anyone would want to still my identity. I don't have money and live a pretty boring life. Well, besides my awesome family and friends.

I need to actually make dinner now instead of just think about it but before I go, Charlie and the Chocolate Factory is coming out soon on IMAX here. I am going to see it and am really looking forward to it. Hmmm, Johnny Depp. Hmmm...

Hello, is it me your looking for...

Because I am in the exact spot you left me. Sorry, I am in a weird mood. I have been online writing people emails and goofing around for about two hours and I have to wait for four more hours before I can leave. I am at work, waiting for packages since my PI is out of town and her post-doc is out of the lab. I don't mind the waiting but I can only read so many scientific papers before my brain leaks out my ears. I read some and then I write an email. I read some, then I take an online test. You get the idea. I do know a lot more about the mechanics of asthma caused by environmental containments in the air. Otherwise known as irritants.

I would summarize everything that I have read but I really want people to come back and read my blog. Of course, that is assuming that people actually read my blog. I could run an experiment on seeing how many people read my blog versus how many people really don't care about me even though each time I see them they say they do. Nah, I'm just kidding. I know that everyone loves me. How could you not? I mean, I am so lovable.

I guess I should probably go since I have been rambling on for a while now and don't think I am actually going to stop anytime soon, which is probably not good news for all the people that I plan on emailing after I post this. You see, my philosophy is that if people don't email me or comment on my blog, that means they must have lost my website, so I should send them reminder messages.

Why can't we all just get along...oh right we're human

I don't have the time or inclination to write a lot right now but I just wanted to make the comment that I am so freakin' tired of waking up to hear about more violence. I understand peoples need to get their voices heard and when it seems that no one is listening, jumping up and down and waving arms seems like the best idea. Isn't that what killing innocent bystanders amounts to? The whinning of a five year old child? I know that it is hard to get people to listen, but I am also sick and tired of people lumping other people in the same category just because they look the same or come from the same place. People are different, that is what makes them interesting. If everyone was the same, it would be boring. Well, either that or crazy. I could not imagine 5 billion Julies walking around. A-no one would talk to each other, B-the human race would die off, and C-books would out sell everything.

Everyone has the right to live a peaceful and full life as long as they do not tread on others.

Lymphocytes migrate through endothelial tissues using pathways mediated by receptors, one of which is the binding of VCAM-1 to α4-integrin of the lymphocyte. The VCAM-1 then stimulates the calcium-and Rac1-dependent activation of NADPH oxidase. The NADPH oxidase produces superoxide that dismutates to H2O2, generating ROS. ROS activates MMPs, which degrade extracellular matrix and cell-cell junctions allowing cell retraction and the migration of lymphocytes.
We have observed the function of NADPH oxidase in vivo by using CYBB mice (gp91phox -/-) and wild type mice. Gp91phox is the catalytic subunit of NADPH oxidase. VCAM-1 dependent infiltration of eosinophils into the lung was induced by OVA intranasally. The mice were then sacrificed, the lungs were lavaged, and then the lungs were frozen for tissue analysis of leukocyte subsets. The tissue samples will be stained for major basic protein-1 (MBP-1) for eosinophil detection and stains for B and T cell surface markers. The tissue samples will then be counted to determine the number of lymphocytes within the tissue and blood vessels. Since the samples were lavaged, few leukocytes will be found in the alveolar cavities.
The importance of this experiment is to show that without NADPH oxidase activity, eosinophils cannot migrate across endothelial cells because VCAM-1 dependent ROS cannot be synthesized. Since ROS cannot be synthesized, MMPs would not be activated and there would be reduced retraction of the cell junctions. This could reduce inflammation caused by eosinophils in allergen-induced asthma.

So for you who are not trained in the biological sciences, this is the summary of what I will be doing this summer in my lab. I wrote it today as my first assignment in the lab. I basically will be counting cells in lung tissue slides on the microscope. I will also be helping the lab do bleeds for mice and at the end of the summer I will learn how to do western blots, which I don't understand enough right now to explain. It is very exciting and I can't wait to actually start it all.
